Xanthurenic Acid 8-O-&~-Glucoside, a Novel Tryptophan Metabolite in Eye-color Mutants of Drosophila meZanogaster*

چکیده

An unknown fluorescent metabolite has been isolated from heads of eye-color mutants of Drosophila melanogaster. Only a few mutations cause it to accumulate, uiz. cardinal (cd), dark red brown (drb), Henna-recessive (Hn’), purple (pr), Punch2 (Pu’), Punch-Grape (Puff’), and scarlet (st). After purification by ion-exchange chromatography, the spectroscopic, chemical, and enzymatic analyses revealed that it is a novel quinoline derivative: xanthurenic acid 8-0-,9-D-glucoside. Feeding experiments suggest that this glucoside is synthesized from 3-hydroxykynurenine and that free xanthurenic acid is not a precursor. The results from the analysis for its occurrence in double mutants, together with the fact that xanthurenic acid 8-glucoside share the same precursor as xanthurenic acid and xanthommatin, suggest that xanthurenic acid 8-glucoside formation is closely related to the regulation of the last step in the biosynthesis of xanthommatin.

Tryptophan metabolite excretion by the American cockroach.

Abstrac t -1 . Three fluorescing materials have been discovered in excreta from the American cockroach, Periplaneta americana (L.), and found to increase in concentration as dietary nitrogen increases. 2. These compounds have been identified as kynurenic, xanthurenic and 8-hydroxyquinaldic acids. 3. Feeding cockroaches antibiotic eliminates 8-hydroxyquinaldic acid production, presumably by inhi...

Identification of xanthurenic acid 8-O-beta-D-glucoside and xanthurenic acid 8-O-sulfate as human natriuretic hormones.

Hormonal regulation of salt excretion and water balance by the kidneys is well documented. Before 1961, it was widely believed that the glomerular filtration rate and the steroid hormone aldosterone controlled sodium balance in the body. In 1961, deWardener et al. [de Wardener HE, Mills IH, Clapham WF, Hayter CJ (1961) Clin Sci 21:249-258] showed that when these two variables were controlled, t...

Renal effects of a novel endogenous natriuretic agent xanthurenic acid 8-o-β-d-glucoside in rats

Xanthurenic acid 8-o-β-d-glucoside is an endogenous derivative of tryptophan metabolism, isolated from urine of patients with chronic renal disease. This compound was suggested previously to act as a natriuretic hormone based on its ability to block short circuit currents in a frog skin assay and to induce a sustained natriuresis when injected into rats (C. D. Cain et al., Proc. Natl. Acad. Sci...

Diuresis by intravenous administration of xanthurenic acid in rats, and inhibition by probenecid.

The conjugates with sulfate and glucoside of xanthurenic acid, a tryptophan metabolite, were reported to show natriuresis. Sulfotransferase for xanthurenic acid works in the renal proximal tubule to produce the sulfate of xanthurenic acid as well as the liver, and we recently found that xanthurenic acid is a substrate of renal organic anion transporter OAT1. The purpose of this study was to exa...
